測(cè)定意義
UDPG焦磷酸化酶是生物體糖原合成過(guò)程中的關(guān)鍵酶。在葡萄糖合成糖原前催化葡萄糖活化，將1-磷酸葡萄糖與UTP分子合成為UDP-葡萄糖（UDPG）。
測(cè)定原理
UGP可逆催化反應(yīng)生成1磷酸葡萄糖，在磷酸葡萄糖變位酶和6-磷酸葡萄糖脫氫酶作用下將NADP轉(zhuǎn)化為NADPH，340nm的吸光值增加速率反映了UGP活性。
自備實(shí)驗(yàn)用品及儀器
天平、低溫離心機(jī)、研缽、紫外分光光度計(jì)/酶標(biāo)儀、微量石英比色皿/96孔板。

（1）請(qǐng)參照相關(guān)法規(guī)、文獻(xiàn)、MSDS等信息，理解并掌握好試劑的特性，再進(jìn)行安全操作。
（2）通常購(gòu)買(mǎi)試劑時(shí)一定要想到一次性用完。若估算不足有剩余的話，更加注意其他保管和管理。
（3）萬(wàn)一試劑操作者并非專(zhuān)業(yè)技術(shù)人員。必須在專(zhuān)業(yè)人士的指導(dǎo)監(jiān)督下進(jìn)行操作。對(duì)于使用后的廢棄物，不要或者舊的試劑，應(yīng)按照相關(guān)法律法規(guī)正當(dāng)處理。
（4）購(gòu)買(mǎi)后，請(qǐng)務(wù)必確認(rèn)標(biāo)簽上的注意事項(xiàng)；做好預(yù)防顛倒，摔壞的措施和管理體制；在使用前再次確認(rèn)標(biāo)簽上的注意事項(xiàng)，做好必要的安全對(duì)策；對(duì)沒(méi)有標(biāo)明其危害特性、有害特性的，也要慎重地進(jìn)行操作；必須帶好防護(hù)用具，小心翼翼進(jìn)行操作。
樣本處理方法：
1、血清（漿）樣品：直接檢測(cè)。
2、細(xì)菌、細(xì)胞或組織樣品的制備： 
細(xì)菌或培養(yǎng)細(xì)胞：先收集細(xì)菌或細(xì)胞到離心管內(nèi)，離心后棄上清；按照細(xì)菌或細(xì)胞數(shù)量
（104個(gè)）：提取液體積（mL）為500~1000：1 的比例（建議 500萬(wàn)細(xì)菌或細(xì)胞加入 1mL提取液） ，超聲波破碎細(xì)菌或細(xì)胞（冰浴，功率20％或200W，超聲 3s，間隔10s，重復(fù)30 次）8000g 4℃離心 10min，取上清，置冰上待測(cè)。 
組織：按照組織質(zhì)量（g） ：提取液體積(mL)為 1：5~10 的比例（建議稱(chēng)取約 0.1g組織，
加入 1mL提取液），進(jìn)行冰浴勻漿。8000g 4℃離心10min，取上清，置冰上待測(cè)。
