細(xì)胞膜鈣ATP酶4B抗體,Anti-PMCA4B抗體背景介紹：The protein encoded by this gene belongs to the family of P-type primary ion transport ATPases characterized by the formation of an aspartyl phosphate intermediate during the reaction cycle. These enzymes remove bivalent calcium ions from eukaryotic cells against very large concentration gradients and play a critical role in intracellular calcium homeostasis. The mammalian plasma membrane calcium ATPase isoforms are encoded by at least four separate genes and the diversity of these enzymes is further increased by alternative splicing of transcripts. The expression of different isoforms and splice variants is regulated in a developmental, tissue- and cell type-specific manner, suggesting that these pumps are functionally adapted to the physiological needs of particular cells and tissues. This gene encodes the plasma membrane calcium ATPase isoform 4. Alternatively spliced transcript variants encoding different isoforms have been identified.

保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

細(xì)胞膜鈣ATP酶4B抗體溶解方法:

方法1：我們的抗體（凍干粉）已經(jīng)包含了0.01M PBS、1% BSA和0.1% 防腐劑（疊氮鈉或*），您只需要加入滅菌的雙蒸水就行了?？贵w溶解后，置于-20℃保存，分裝后使用，避免反復(fù)凍融，可保證至少1年有效；
方法2：也可以只加入一半體積的雙蒸水，再用一半體積的甘油補(bǔ)足，置于-20℃保存，這樣抗體不會(huì)凍，有利于抗體的穩(wěn)定。
后續(xù)試驗(yàn)時(shí)，再使用對(duì)應(yīng)的緩沖液稀釋成工作液，即用即配。
我們的抗體（凍干粉），在常溫放置下，至少一個(gè)月有效；若在-20℃下保存（*），至少三年有效。
Antibodies are prepared with 0.01M PBS, pH 7.4 with 1% BSA and 0.1% Sodium Azide, prior to lyophilization. Currently, we offer both lyophilized and reconstituted antibodies, both resulting in final concentration of 1mg/mL. We recommend two ways to reconstitute antibodies in lyophilized form:
1) Reconstituting by directly adding 100uL of sterile, distilled water.
2) Alternatively, if glycerol does not affect the experiment, add 50uL sterile distilled water, followed by adding 50uL of glycerol.

3.水化。依次經(jīng)過(guò)無(wú)水乙醇、95%、90%、80%、70%，各5分鐘，蒸餾水（或自來(lái)水）5分鐘
4.PBS洗5分鐘，三次
5.抗原修復(fù)。枸櫞酸緩沖液（約92-95℃）煮沸（沸水?。┬迯?fù)15分鐘，自然涼至室溫。PBS洗5分鐘，三次
6.3%雙氧水消化內(nèi)源性的過(guò)氧化物酶，20分鐘，PBS洗5分鐘，三次
7.正常山羊血清封閉，37℃20分鐘
8.一抗孵育，4℃過(guò)ye.
9.PBS洗5分鐘，三次
10.二抗孵育，37℃20分鐘（二抗為*標(biāo)記的羊抗兔IgG），PBS洗5分鐘，三次
11.三抗孵育，37℃20分鐘（三抗為辣根酶標(biāo)記的鏈霉親和素），PBS洗5分鐘，三次
12.DAB顯色，鏡下觀察結(jié)果，適時(shí)終止
13.蘇木素復(fù)染5分鐘，自來(lái)水沖洗片刻，1%的鹽酸酒精（75%）溶液（請(qǐng)自行配置，配制比例為：100ml 75%酒精+1ml濃鹽酸）分化30秒，自來(lái)水沖洗5分鐘藍(lán)化
14.脫水。依次經(jīng)過(guò)70%、80%、90%、95%、無(wú)水乙醇，各5分鐘，二甲苯15分鐘，三次
15.封片，中性樹(shù)膠封片
