Elisa實驗常用方法： 
雙抗體夾心法測定抗原 

將抗原免疫*種動物獲得的抗體吸附于固相表面;

加抗原，形成抗原-抗體復(fù)合物; 

加抗原免疫第二種動物獲得的抗體，形成抗體抗原抗體復(fù)合物;

加酶標(biāo)抗抗體(第二種動物抗體的抗體); 

加底物。底物的降解量＝抗原量。

Elisa實驗原理：酶結(jié)合物與相應(yīng)抗原或抗體結(jié)合后，可根據(jù)加入底物的顏色反應(yīng)來判定是否有免疫反應(yīng)的存在，而且顏色反應(yīng)的深淺是與標(biāo)本中相應(yīng)抗原或抗體的量成正比例的，所以，可以按底物顯色的程度顯示試驗結(jié)果。
