辣根過(guò)氧化物酶（HRP）是一種糖蛋白，每個(gè)分子含有一個(gè)氯化血紅素（protonhemin）區(qū)作輔基。酶的濃度和純度常以輔基的含量表示。氯化血紅素輔基的zui大吸收峰是403nm，HRP酶蛋白的zui大吸收峰是275nm，所以酶的濃度和純度計(jì)算式是（已知HRP的A（1cm 403nm 1%）＝25，式中1％指HRP百分濃度為100ml含酶蛋白1g，即10mg/ml，所以，酶濃度以 mg/ml 計(jì)算是HRP的A（1cm 403nm mg/ml=2.5）HRP純度（RZ）=A403nm/A275nm純度RZ（Ｒeinheit Zahl）值越大說(shuō)明酶內(nèi)所含雜質(zhì)越少。高純度HRP的RZ值在3.0左右，zui高可達(dá)3.4。用于ELISA檢測(cè)的HRP的RZ值要求在3.0以上。

ELISA的基本原理有三條： 
    （1）抗原或抗體能以物理性地吸附于固相載體表面，可能是蛋白和聚苯乙烯表面間的疏水性部分相互吸附，并保持其免疫學(xué)活性；
    （2）抗原或抗體可通過(guò)共價(jià)鍵與酶連接形成酶結(jié)合物，而此種酶結(jié)合物仍能保持其免疫學(xué)和酶學(xué)活性； 
    （3）酶結(jié)合物與相應(yīng)抗原或抗體結(jié)合后，可根據(jù)加入底物的顏色反應(yīng)來(lái)判定是否有免疫反應(yīng)的存在，而且顏色反應(yīng)的深淺是與標(biāo)本中相應(yīng)抗原或抗體的量成正比例的，因此，可以按底物顯色的程度顯示試驗(yàn)結(jié)果。
