辣根過(guò)氧化物酶（HRP）是一種糖蛋白，每個(gè)分子含有一個(gè)氯化血紅素（protonhemin）區(qū)作輔基。酶的濃度和純度常以輔基的含量表示。氯化血紅素輔基的zui大吸收峰是403nm，HRP酶蛋白的zui大吸收峰是275nm，所以酶的濃度和純度計(jì)算式是（已知HRP的A（1cm 403nm 1%）＝25，式中1％指HRP百分濃度為100ml含酶蛋白1g，即10mg/ml，所以，酶濃度以 mg/ml 計(jì)算是HRP的A（1cm 403nm mg/ml=2.5）HRP純度（RZ）=A403nm/A275nm純度RZ（Ｒeinheit Zahl）值越大說(shuō)明酶內(nèi)所含雜質(zhì)越少。高純度HRP的RZ值在3.0左右，zui高可達(dá)3.4。用于ELISA檢測(cè)的HRP的RZ值要求在3.0以上。

elisa實(shí)驗(yàn)原理
    用于免疫酶技術(shù)的酶有很多，如過(guò)氧化物酶，堿性磷酸酯酶，β－Ｄ－半乳糖苷酶，葡萄糖氧化酶，碳酸酐酶，乙酰*酯酶，6－磷酸葡萄糖脫氧酶等。常用于ELISA法的酶有辣根過(guò)氧化物酶，堿性磷酸酯酶等，其中尤以辣根過(guò)氧化物酶為多。由于酶摧化的是氧化還原反應(yīng)，在呈色后須立刻測(cè)定，否則空氣中的氧化作用使顏色加深，無(wú)法準(zhǔn)確地定量。
