檢測(cè)原理：采用雙抗體夾心ABC-ELISA法

Elisa試劑盒實(shí)驗(yàn)原理：用純化的抗體包被微孔板，制成固相載體，往包被抗LEP抗體的微孔中依次加入標(biāo)本或標(biāo)準(zhǔn)品、*化的抗LEP抗體、HRP標(biāo)記的親和素，經(jīng)過(guò)*洗滌后用底物TMB顯色。TMB在過(guò)氧化物酶的催化下轉(zhuǎn)化成藍(lán)色，并在酸的作用下轉(zhuǎn)化成zui終的黃色。顏色的深淺和樣品中的LEP呈正相關(guān)。用酶標(biāo)儀在450nm波長(zhǎng)下測(cè)定吸光度（OD值），計(jì)算樣品濃度

樣品類型（SAMPLE TYPES）：標(biāo)本必須為液體，不含沉淀。包括血清、血漿、尿液、胸腹水、腦脊液、細(xì)胞培養(yǎng)上清、組織勻漿等。1ml的全血可得到0.5ml的血清或血漿。每個(gè)標(biāo)本量收集體積＝100ul×檢測(cè)種類。取材前須向銷售人員索要說(shuō)明書(shū)。 

樣品采集：收集標(biāo)本前必須清楚要檢測(cè)的成份是否足夠穩(wěn)定。對(duì)收集后當(dāng)天進(jìn)行檢測(cè)的標(biāo)本，儲(chǔ)存在4℃?zhèn)溆?，如有特殊原因需要周期收集?biāo)本，將標(biāo)本及時(shí)分裝后放在-20℃或-70℃條件下保存。避免反復(fù)凍融。標(biāo)本2-8℃可保存48小時(shí)，-20℃可保存1個(gè)月。-70度可保存6個(gè)月。部分激素類標(biāo)本需添加*。
